Today we put our mutated fragment of DNA and the big plasmid fragment called Prk together to ligate. This will make a ring of plasmid with Prk and small mutated fragment together as one. this plasmid will be transformed IN A S17 E.COLI BACTERIUM. I am very happy to reach till this point as people with “m” subunit are having problem with lysis.

Yesterday we went to robotics competition and we were amazed to see students involvement as well as enthusiasm. These days robotics has become somewhat simple with already available commands which one can drag and drop to create their own robot. Though students need to understand how they would like their robot to perform and based on that they have to logically sequence their chosen commands. Finally the robot will be monitored by computer to carry out its functions.

Today we practised electrophorasis gel run. It was fun :) Our L submit went well but there is some problem with M subunit. It seems like the enzyme did not cut the plasmid as calculated. May be the enzyme has denatured or.......We will find more about it tomorrow.

Today we were trying to develop leson plan which can integrate all the subjects we taught in our group. We decided that for chemistry and bilogy class we can do part of our research activity followed by lab report. Lab report will be based on the outline given to them which will challege their resoning capabilities. For math class they can calculate the cost of small scale lab set-up on Excel Spreadsheet.
